Simultaneous Production and Immobilization of Lipase Using Pomegranate-Seed Residue: A New Biocatalyst for Hydrolysis Reactions and Structured Lipids Synthesis

Pomegranate-seed residue (PSR) was used in a new strategy for the simultaneous production of Yarrowia lipolytica lipase by submerged fermentation and its immobilization adsorption. This biocatalyst—the fermented solid containing adsorbed (fermPSR)—was evaluated hydrolysis reactions structured lipid synthesis. In shake flasks, yeast extract urea were best nitrogen sources with PSR their use increased even further. result confirmed 3.5-liter bioreactor, activity an extracellular medium 40 U/mL. A maximum reaction rate (Vmax) 49.5 µmol/min/g, Michaelis–Menten constant (Km) 207 µmol/L, turnover number (Kcat) 130 s−1 determined biocatalyst, fermPSR, p-nitrophenyl laurate (p-NPL) into p-nitrophenol. The conversion p-NPL p-nitrophenol subsequent fermPSR’s potential industrial hydrolytic reactions. lipids from vegetable oil free fatty acids fermPSR evidences versatility this biocatalyst.
